摘要 目的研究EphB4受体和缺氧诱导因子-1α(HIF-1α)蛋白在胃癌组织中的表达,并探讨其生物学意义及相互关系。方法应用免疫组化法检测74例胃癌组织(胃癌组)和13例正常胃黏膜组织(对照组)中EphB4、HIF-1α的表达,并分析其表达与胃癌临床病理因素之间的关系。结果胃癌组中EphB4和HIF-1α的高表达率分别为62.16%(46/74)和52.70%(39/74),在对照组中,EphB4和HIF-1α的高表达率分别为15.38%(2/13)和0%(0/13)。胃癌组织中EphB4和HIF-1α的表达显著高于正常胃黏膜组织(P<0.01)。EphB4、HIF-1α的表达与胃癌的组织学分级及临床病理分期关系密切(P<0.05),而与年龄、性别、肿瘤直径、淋巴结转移、远处转移无关(P>0.05)。与HIF-1α不同,EphB4的表达还与肿瘤浸润深度有关(P<0.05)。此外,EphB4与HIF-1α的表达成显著正相关(r=0.2091,P<0.01)。结论在胃癌中EphB4、HIF-1α均高表达,其表达程度与胃癌的恶性程度有关,而且EphB4表达与HIF-1α的表达呈正相关。过表达的EphB4、HIF-1α在胃癌恶性进展中发挥重要作用。 Objective To investigate the expressions of EphB4 receptor and hypoxia inducible factor 1α (HIF-1α) protein in human primary gastric carcinoma, and to study the biological implication of their expression. Methods The expressions of EphB4 receptor and HIF-1α protein were detected in 74 specimens of human primary gastric carcinoma (gastric carcinoma group) and in 13 normal gastric mucosa tissues (control group) by immunohistochemistry. Correlations between the expression of these proteins, and clinical and pathological features were respectively analyzed. Result High expressions of EphB4 receptor and HIF-1α protein were found to be 62.2% (46/74) and 52.7% (39/74) respectively in gastric carcinoma group, while the expressions of EphB4 receptor and HIF-1α protein in control group were 15.38% (2/13) and 0% (0/13), respectively. The differences were significant between two grrups (P〈0.01). In gastric carcinoma group, the expressions of EphB4 receptor and HIF-1α protein were closely related to the pathological grades and clinical stages of gastric carcinoma (P〈0.05), but there was no correlation with age, gender, tumor diameter, lymph node metastasis and distant metastasis. The expression of EphB4 receptor, which was different from HIF-1α protein, was also related to the depth of gastric carcinoma invasion (P〈0.05). In addition, a significant positive correlation was observed between EphB4 receptor and HIF-1α protein expression (r=0.209 1, P〈0.01). Conclusion It is found that in gastric carcinoma, both EphB4 receptor and HIF-1α protein are highly expressed, and the degree of expression is correlated with malignant degree of gastric carcinoma. The expression of EphB4 receptor show a positive correlation with the expression of HIF-1α protein. The higher expressions of EphB4 receptor and HIF-1α protein play an important role in the malignant development of gastric carcinoma.
