抗抑郁药物对海马神经元保护作用的研究进展 被引量:20

摘要 就目前研究来说,海马的体积进行性缩小、结构改变、神经元丢失以及成年个体海马齿状回神经发生的降低都与抑郁症特别是严重抑郁症有很大的关联。因此,有人认为抗抑郁药物可能是通过保护海马神经元的死亡,促进齿状回的神经发生发挥其主要作用的。本综述中主要探讨2个问题:一是回顾以前的临床前及临床研究,提出抑郁症导致海马体积变小与细胞变化的主要证据;二是讨论长期的抗抑郁药治疗通过影响谷氨酸受体、脑源性神经营养因子的表达、5-羟色胺受体及清除自由基来阻止海马神经元损伤的可能机制。 The hippocampus is a brain structure involved in depression, especially in major depression, in which a progressive atrophy of the hippoeampal formation, accompanying hippoeampus structural reorganization, neuron loss, and decrease of neurogenesis in dentate gyrus granule of adult animals was reported. It is possible that the most of antidepressants produced the antidepression effects by neurons protective effect in hippoeampus or pro- moting neurogenesis in dentate gyrus. Two issues were discussed in the review. Firstly, the evidences of the hippo- campus changes, including structure and cellular changes in preelinieal and clinical research, were reviewed. Sec- ondly, the probable mechanisms that chronic antidepressants treatment reduced hippoeampus neurons injury by the influence on the Glu receptor, BDNF expression, 5-HT receptor and clean of the free radical, were discussed.
