摘要
目的:研究红景天提取物对大鼠心肌缺血再灌注损伤后血清细胞因子水平的影响。方法:SD大鼠灌胃红景天提取物5 g.kg-1.d-1,qd,连续5 d。末次给药后用冠状动脉结扎法制备大鼠I/R模型,再灌注60 min后处死大鼠,取血,用放射免疫分析方法检测血清内皮素(ET)、肿瘤坏死因子-α(TNF-α)和白介素-6(IL-6)含量。实验同时设假手术组和苯那普利阳性对照组(10 g.kg-1.d-1)。结果:与假手术组比较,模型组血清TNF-a,IL-6和ET水平均显著增加(P<0.01)。与模型组比较,红景天提取物组血清TNF-α,IL-6和ET均显著降低(P<0.05),但仍高于假手术组(P<0.01);红景天提取物组与苯那普利组比较,无统计学差异。结论:红景天提取物对心肌缺血再灌注损伤有明显的保护作用,作用机制可能与其降低血中细胞因子水平有关。
Objective: To investigate the effects of extracts from Hongjingtian on cytokines after myocardial ischemia and reperfusion injury (MIRI). Methods : After administration of Hongjingtian extracts (5 g· kg^- 1· d^ - 1, qd) for 5 days, the rats were subjected to ligation of the left anterior descending coronary arteries for 60 minutes and reperfusion for 60 min to induce MIRI. The serum ET, TNF-α and IL-6 was determined by radioimmunoassay. Sham operation and benazepril group (positive control, 10 g·kg-α ·d^-1, qd) were used as the controls. Results. After MIRI, the serum ET, TNF-α and IL-6 were remarkably decreased as compared wih sham operation (P 〈 0. 01 ). Hongjingtian extracts markedly lifted the decreased levels of ET, TNF-α and IL-6 in rats with MIRI; the effect was similar to that of benazepril. Conclusion : Hongjingtian extracts exert remarkable protective effects on MIRI. This effect may relate to the decreased levels of serum cytokines.
