摘要
目的:探讨多胺类似物CHEN,CPEN和BENS对宫颈癌细胞株Hela生长的影响。方法:MTT法检测细胞的生长状况;亚凋亡峰流式细胞分析法及DNA片段化分析法检测细胞凋亡;化学分析法测定精胺氧化酶(SMO)活性。结果:3种多胺类似物均可显著抑制Hela癌细胞增殖,抑制作用随药物浓度加大而增加,其细胞毒性大小依次为CHEN>CPEN>BENS。细胞凋亡分析发现,CHEN,CPEN和BENS可诱导Hela细胞程序性凋亡,导致凋亡峰出现和明显的DNA片段化。用10μmol.L-1CHEN,CPEN和BENS处理Hela细胞24 h可显著性抑制精胺氧化酶活性。结论:CHEN,CPEN和BENS通过诱导细胞凋亡而抑制Hela癌细胞生长,其诱导凋亡的作用与精胺氧化酶活性无关。
Objective:To study the effects of polyamine analogues, CHEN [ N^1-ethyl-N^11-( cycloheptyl)- methyl-4,8, diazaundeeane ], CPEN [ N^1 -ethyl-N^11 -( eyclopropyl)-methyl-4,8-diazaundeeane ] and BENS [ his (ethyl) -norspermine ] on the growth of Hela human cervical cancer cells. Methods : MTT reduction was used to assay the cell proliferation. Sub-G1/flow cytometry and DNA fragmentation assays were used to determine the cell apoptosis. Chemical analysis method was used to determine the enzymatic activity of spermine oxidase. Results:All the polyamine analogues inhibited the growth of Hela cancer cells in a concentration-dependent manner with an order of cytotoxicity : CHEN 〉 CPEN 〉 BENS. CHEN, CPEN and BENS induced apoptosis of Hela cells and resulted in appearance of sub-G1 peaks and DNA fragmentations. Significant decrease of spermine oxidase activities were observed after 24-h treatment with 10 μmol · L^-1 CHEN, CPEN and BENS. Conclusion: Polyamine analogues ( CHEN, CPEN and BENS) inhibit proliferation of the Hela cervical cancer cells by inducing apoptosis; the inhibition of apoptosis is not related to the activity of spermine oxidase.
