摘要
目的探讨单个核细胞(PMBC)凋亡是否可以作为评价透析膜生物相容性的指标。方法选择卫生部中日友好医院肾病中心维持性血液透析患者分别用醋酸纤维膜(CA)和聚砜膜(PS)与血液透析患者PMBC孵育不同时间(0、24、48、72h),以流式细胞术、酶联免疫法(ELISA)检测不同透析膜对PMBC凋亡、白细胞介素-1(IL-1)及肿瘤坏死因子(TNF-a)合成的影响。结果①CA膜组PMBC凋亡程度明显高于PS膜及正常对照组(P<0.05),PS膜组与正常对照之间差异无显著性(P>0.05);②对PMBC凋亡OD值和IL-1及TNF-a合成量成正相关(r=0.725;r=0.813,P<0.01)。结论生物相容性不同的透析膜对透析患者PMBC凋亡存在着不同程度的影响,PMBC凋亡程度可以作为衡量透析膜生物相容性的指标。
Objective To study whether apoptosis of peripheral blood mononuclear cells (PMBC) can be used as an index to evaluate the biocmpatibility of hemodialysis membranes. Methods PMBC from hemodialysis patients were incubated with different hemodialysis membranes for 0 to 72 hours. PMBC apoptosis and their interleukin-1 and tumor necrosis factor-a synthesis were evaluated by flow cytometry and ELISA. Results Apoptosis rate was higher in PBMC incubated with cellulose acetate membrane than in those incubated with polysulfone membrane and those without incubation, but the rate had no difference between the PBMC incubated with polysulfone membrane and those without incubation. PBMC Apoptosis was associated with the increase of interleukin-1 and tumor necrosis factor-a in the media. Conclusion PMBC apoptosis can be used as an index to evaluate the biocompatibility of hemodialysis membranes.
