Modification of Bovine Serum Albumin with Polyethylene Glycol

摘要 用三聚氯氰活化的聚乙二醇(PEG-1900和PEG-5000)修饰牛血清白蛋白(BSA)的游离氨基,经透析,冻干,得到PEG修饰的牛血清白蛋白.对PEG修饰的BSA进行了理化、生物学及部分药用指标测定.实验表明,用PEG-1900和PEG-5000修饰时,修饰率分别达到90%和50%左右时,便可消除BSA的过敏反应. PEG-1900 and PEG-5000 were activated by using cyanuric chloride then coupled with BSA to obtain PEG-1900-BSA and PEG-5000-BSA. The physico-chemical, biological, and pharmaceutical properties were studied. The results showed that the modified bovine serum albumin appeared to lose its immunogenicity when the degree of the modification of the amino groups in the bovine albumin molecule was about 90% by using PEG-1900 or was about 50% by using PEG-5000. The best modification. condition was 18 h at 25 ℃. The bovine serum albumin modified by PEG not only eliminated allergic reaction from BSA but also changed its physical and chemical properties. PEG-modified BSA can desolve over a wide ranges of pH, at least from pH 1. 0 to pH 12. 0. Some quality index examinations indicated that it is possible to substitute the PEG-bovine serum albumin for human albumin.
