摘要
苜蓿根瘤菌突变株JH66菌体生长最适培养基为:蔗糖5g,牛肉膏1g,蛋白胨1g,K2HPO40.0.5g,CaSO40.2g,Na2MoO40.01g,H2O1L,pH7.0.培养1d后,补加葡萄糖至聚β-羟基丁酸(PHB)积累最适碳氮比为15:1,继续培养1d,干菌体PHB含量有显著提高.
The Rhizobium melitoti mutant strain JH66 was grown in the optimal medium that contained 5 g sucrose, 1 g beef-extraction, 1 g peptone, 0.5 g K2HPO4, 0. 2g CaSO4, 0. 01 g Na2MoO4 and 1 L H2O at PH 7. 0. After cultured for 1 day, glucose was added to the medium to make the optimal combination between carbon sourse and nitrogen sourse for PHB accumulation. Continued to be cultured for 1 day, the PHB content in dry cells was improved. The PHB molecular weight was between 270-920 thousand.
