加味四逆散对慢性应激模型大鼠下丘脑单胺类神经递质的影响 被引量:9

The Effect of Jiawei Sini Powder on Monoamine Neurotransmitter of Hypothalamus in Rat Chronic Stress Model

摘要 目的:观察加味四逆散对慢性应激模型大鼠下丘脑单胺类神经递质的影响。方法:选择OPEN-FIELD法评分相近的40只成年SD雄性大鼠随机分为对照组、模型组、氟西汀组、加味四逆散小剂量组和加味四逆散大剂量组,每组各8只。应用分养和长期不可预见的中等强度应激造成大鼠慢性应激抑郁状态。以HPLC-EC法对照研究大鼠下丘脑单胺类神经递质的改变。结果:与对照组比,模型组大鼠下丘脑NE升高,5-HIAA和5-HT降低。与模型组比,氟西汀组、加味四逆散组NE降低而5-HIAA和5-HT升高。结论:慢性应激模型大鼠下丘脑单胺能神经功能活动失常,以去甲肾上腺素的升高和5-羟色胺的下降为主,加味四逆散的作用机理可能与平衡下丘脑单胺能神经的功能状态有关。 Objective:To investigate the effect of Jiawei Sini Powder on monoamine transmitters of Hypothalamus of rat chronic stress model. Methods:Male Sprague-Dauley rats were randomly allocated to five groups as follows:control, model, fluoxetine, Jiawei Sini Powder with small dose and largt dose. The model was produced by sepration and chronic unpredictable mild stress. The behavior was detected by open-field test. HPLC-EC was used to study the changes of monoamine transmitters of Hypothalamus. Results:The results showed that NE level of model group is higher, and 5-HIAA and 5-HT are lower. Fluoxetine and Jiawei Sini Powder decreased the NE level and increased 5-HIAA and 5-HT. Conclusions: The data suggested that dysfanction of monoamine nerve of hypothalamus in rat chronic stress model is related with increased NE and decreased 5-HT, regulation for which may explain the mechanism of Jiawei Sini Powder.
