摘要
目的慢性支气管炎患者在使用阿奇霉素氯化钠注射液前后,分析外周血中杀菌/通透性增加蛋白(Bactericidal/permeability Increasing protein,BPI)mRNA的表达水平,探讨BPI在炎症过程中的作用机制。方法应用巢式RT-PCR技术,检测22例慢性支气管炎患者和18例健康志愿者外周血中BPImRNA的表达水平,以β-actin为内标计算其相对表达量,并运用SPSS10.0统计软件对半定量结果进行t检验。结果18例健康受试者外周血中BPImRNA的平均相对表达水平为(0.74±0.40);22例慢支患者在使用阿奇霉素氯化钠注射液治疗前后,外周血中BPImRNA的平均相对表达水平分别为(0.46±0.23)和(0.24±0.19);经t检验,健康对照组与慢支患者,以及慢支患者用药前后,其外周血中BPImRNA的相对表达水平均有显著性差异(P<0.05)。结论慢性支气管炎患者在使用阿奇霉素氯化钠注射液治疗后外周血BPImRNA的表达水平较低;BPI表达水平下降,体内自身清除细菌能力下降。
Objective To analyze the expression of bactericidal/permeability increasing protein (BPI) mRNA in peripheral blood of patients with chronic bronchitis (CB) before and after Azithromycin Sodium Chloride Injection was used, and investigate the role of BPI on the inflammatory treatment. Methods Nested PCR was used to detect BPI mRNA expression in peripheral blood from 22 patients with CB and 18 normal healthy volunteers. The relative expression amount of BPI was calculated through β -actin as a internal standard. The semi -quantitative results were analyzed by Student's t - test using SPSS10.0. Results The relative expression levels of BPI mRNA in peripheral blood of 18 normal healthy volunteers were(0. 74 ± 0. 40). The relative expression levels of BPI mRNA in peripheral blood of 22 patients with CB before and after Azithromycin Sodium Chloride Injection was used were (0. 46 ± 0. 23 ) and (0. 24 ± 0. 19 ) respectively. Among the healthy volunteers and the patients with CB before and after Azithromycin Sodium Chloride Injection was used, the relative expression quantity is different by statistic ( P 〈 0. 05 ). Conclusion The expression levels of BPImRNA in peripheral blood of patients with CB were lower after Azithromycin Sodium Chloride Injection was used, indicating that BPI's antibacterial ability as well as body's immunological competence decreased after antibiotics were used in inflammatory process.
