摘要
本实验以10个入选组多重的引物作为研究对象,探讨了适用于玉米DNA指纹库构建的多重PCR复合扩增体系的建立,分析了影响多重PCR组合建立的主要因素,包括引物扩增片段范围,引物间的相互作用,引物间主带与非特异带的相互影响,并提出了构建多重PCR扩增体系应该遵循的重要原则。
In this research we attempted to established the multiplex SSR-PCR system that might be suitable for maize DNA fingerprinting constructions. Ten candidate primers and six leading maize inbreeding lines were employed to explore the mainly factors which would possible influence on constructing multiplex PCR combination, such as the amplifying range of the primer combinations, the interaction among the primers, the influence between the specific band and non-specific band among the primers. Then we had some suggestions for establishing multiplex PCR combination.
