弥漫性脑损伤模型大鼠细胞因子在额叶表达及血清含量变化

Change in the expression of interleukin-6 and tumor necrosis factor-α mRNA in frontal lobe and in protein level in serum in diffused traumatic brain injury rat

摘要 目的:研究白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α)在弥漫性脑损伤大鼠额叶的表达和血浆含量的变化。方法:采用Mamarou的弥漫性脑损伤的动物模型,分别于伤后不同时间收集额叶脑组织和静脉血,分别以RT-PCR法和放免法测定脑组织中IL-6和TNF-α的mRNA和血清中IL-6和TNF-α的含量变化。结果:在弥漫性脑损伤的额叶脑组织中IL-6 mRNA的表达较对照组明显升高,并在损伤后6h达到高峰;血清中IL-6的表达也较对照组升高,在12h达到高峰。而TNF-α mRNA的表达在12h明显高于对照组,24h达到高峰;血清中的TNF-α水平在24h内逐渐升高。结论:在弥漫性脑损伤中,IL-6和TNF-α在脑组织和血清中升高,在疾病的发展中起到重要的作用。 Objective:To investigate the change in the expression of interleIukin-6(IL-6) and tumor necrosis factor-α(TNF-α)mRNA in frontal lobe and serum IL-6 and TNF-α level in diffused traumatic brain injury rat model.Methods:Mamarou rat model of diffused traumatic brain injury was used in the study.Venous blood and frontal lobe were collected at different time after injury.The change of IL-6 and TNF-α mRNA expression and serum IL-6 and TNF-α level were detected with RT-PCR method and radioimmunoassay,respectively.Results:it was found that IL-6 mRNA level increased in diffused traumatic brain injury compared to the control group and reached to the peak 6 hours after injury.Serum IL-6 level also elevated in diffused traumatic injury rat compared to the control group and reached to the peak 12 hours after injury.TNF-α mRNA expression was up-regulated compared to the control group and reached to the peak 12 hours after injury.Serum TNF-α level elevated gradually within 24 hours after injury.Conclusion:Diffused traumatic brain injury induces upregulation of IL-6 and TNF-α mRNA expression in frontal lobe and elevation of serum level of them.IL-6 and TNF-αmay play an important role in the progress of diffused traumatic brain injury.
