摘要
目的:探讨单纯紫外线(UV)暴露与砷(As)和紫外线联合暴露间在色素代谢方面有无差异。方法:采用体外细胞培养法将细胞分为9组(As:0,0.1,1μmol/L;UV:5,10mJ/cm2;As+UV:0.1μmol/L+5mJ/cm2,1μmol/L+5mJ/cm2,0.1μmol/L+10mJ/cm2,1μmol/L+10mJ/cm2),UV照射后,培养72h,测定黑色素水平、酪氨酸酶(Tyr)活力及TyrmRNA表达水平。结果:10mJ/cm2UV组、0.1μ(mol/L+10mJ/cm2组及1μmol/L+10mJ/cm2组的黑色素水平分别显著高于5mJ/cm2组、0.1μmol/L+5mJ/cm2组、1μmol/L+5mJ/cm2组及对照组;各组Tyr活力及mRNA表达水平与对照组相比无显著差异。结论:单纯UV作用对黑色素产生水平的影响与亚砷酸纳(NaAsO2)和UV联合作用对黑色素产生水平的影响无差异,砷对紫外线的促黑素化作用没有协同效应。
Objective:To study the difference between UV exposure and co-exposure to(UV) and arsenic in their effects on melanin metabolism.Methods:Human melanoma cells G361 were cultivated in vivtro and divided into 9 groups as follows:As:0,0.1,1 μmol/L;UV:5mJ/cm2,10mJ/cm2;As+UV:0.1 μmol/L+5mJ/cm2,1 μmol/L+5mJ/cm2,0.1 μmol/L+10mJ/cm2 and 1 μmol/L+10mJ/cm2,which were then exposed to UV and cultured for another 72 h.Melanin levels,TYR activity and the mRNA expression of TYR were determined thereafter.Results:Melanin levels in groups of 10mJ/cm2 UV,0.1 μmol/L+10mJ/cm2 and 1 μmol/L+10mJ/cm2 were significantly higher than those in groups of 5mJ/cm2 UV,0.1 μmol/L+5mJ/cm2 and 1 μmol/L+5mJ/cm2 and the control group.No significant differences have been found in 9 groups in TYR activity and the expression of mRNA compared with the control group.Conclusions:No significant differences have been found in the effects of UV exposure and co-exposure to UV and arsenic on melanin production level,that is,arsenic has no synergistic effect with UV on melanin metabolism.
