摘要
[目的]研究纳豆芽孢杆菌在对健康和鳗弧菌感染牙鲆肠粘液粘附中的作用。[方法]应用5 mol/L LiCl提取纳豆芽孢杆菌表面蛋白,利用蛋白印迹法鉴定在纳豆芽孢杆菌表面蛋白中分子量及参与粘附的特异蛋白。[结果]对提取的纳豆芽孢杆菌表面蛋白进行SDS-PAGE后,发现仅出现一条明显的主要蛋白条带,分子量为29.58 kDa。经蛋白印迹分析,该蛋白质参与了健康和鳗弧菌感染牙鲆肠粘液的粘附过程;纳豆芽孢杆菌全菌蛋白在健康和鳗弧菌感染牙鲆肠粘液中有一个相同的粘附受体,蛋白分子量为13.91 kDa;健康个体还有一个粘附受体,分子量是29.86 kDa。[结论]纳豆芽孢杆菌的表面蛋白在对牙鲆肠粘液的粘附过程中起着重要作用。
[Objective]The purpose was to study the adhesion action of Bacillus natto to healthy and Vibrio anguillarum-infected intestinal mucus of Paralichthys ofivaceus.[Method]The surface proteins of B.natto were extracted by 5 mol/L Iithium chloride(LiCl).The molecular weights of surface proteins and the specific proteins participated in the adhesion were identified by Western-blotting method.[Result]When the extracted surface proteins of B.natto were made through SDS-PAGE,it was found that only one obvious main protein band appeared,with the molecular weight of 29.58 kDa.Western-blotting analysis showed that this protein participated in the course of adhesion to healthy and V.anguillarum-infected intestinal mucus of P.ofivaceus.The whole proteins in B.natto were in healthy and V.anguillarum-infected intestinal mucus of P.ofivaceus had one same adhesion receptor and its molecular weight was 13.91 kDa.There was also one same adhesion receptor in healthy individual,with the molecular weight of 29.86 kDa.[Conclusion]The surface proteins of B.natto played the important role in adhesion course of intestinal mucus of P.ofivaceus.
