摘要 口蹄疫病毒的146S抗原是口蹄疫的主要免疫性抗原,对该抗原进行快速准确的定量对口蹄疫疫苗质量监测、新型疫苗开发和抗原研究有重要的意义。本研究利用酶联免疫吸附实验快速敏感的特性建立了定量146S抗原的检测方法,证明抗原含量和OD值表示的ELISA结果有对数线性关系,相关系数为0.98。实验建立了计算机自动计算程序,可以利用统计学方法快速计算出抗原含量,ELISA进行146S抗原定量的方法使用方便,结果准确,具有较高的实用价值。 The 146S antigen is intact particles of foot-and-mouth disease (FMD) virus and a main component that induces neutralization antibodies against FMDV. The level of neutralization antibodies is related to the amount of 146S antigen in vaccine, Therefore it is important to quantitate the 146S antigen quickly for vaccine development. In this study, an ELISA assay was deve- loped for rapid quantification the 146S antigen. We found that the amount of 146S antigen had a linear relationship with the logarithtn of OD result from ELISA with a correlation coefficient of 0.98, We also developed a compute program to calculate the amounts of 146S antigen from the result of ELISA. Compared with the ultra centrifugation method, the ELISA method is more reproducible with less than 2 μg variations.
