摘要
目的研究桃红四物汤含药血清对血管平滑肌细胞迁移及β3整合素、MMP9表达的影响,并探讨其相关机制。方法制备桃红四物汤含药血清,体外分离培养大鼠颈总动脉血管平滑肌细胞,将血管平滑肌细胞分为对照组(正常大鼠血清培养)与实验组(桃红四物汤含药血清干预)分别进行处理,采用改良的Boyden chamber观察两组血管平滑肌细胞迁移的活性,采用免疫细胞化学方法检测各组血管平滑肌细胞的β3整合素和MMP9表达的情况。结果对照组血管平滑肌细胞存在迁移,在含药血清干预后,大鼠血管平滑肌细胞迁移细胞数减少、迁移距离缩短(P<0.01),且β3整合素和MMP9的表达均降低(P<0.01)。结论桃红四物汤含药血清可抑制血管平滑肌细胞的迁移,且对β3整合素和MMP的表达均产生负性影响,此影响可能参与了对血管平滑肌细胞的迁移及增殖的抑制作用。
[Objective] To research the effects of blood serum containing Decoction of Four Drugs with Taohong on vascular smooth muscle cells (VSMC) migration and the expression of integrin β3, matrix metalloproteinase9 (MMPg), and to investigate the mechanisms about it. [Methods] Blood serum containing Decoction of Four Drugs with Taohong from rats were prepared. The vascular smooth muscle cells were isolated from common carotid artery of rats and cultured in vitro. Vascular smooth muscle cells were disposed to be cultured with normal blood serum (control group) and blood serum containing Decoction of Four Drugs with Taohong(P 〉0.05). Cell migration activity of each group was assayed by modified micro-Boyden chamber. The expression of integfin β3 and MMP9 of each group were detected by immunocytochemistry staining. [Results] Cell migration existed in normal control group. After intervention with blood serum containing drugs, the migrating cell population diminish and the migration lenth decurtated (P 〈0.01) and the expression of integrin β3 and MMP9 decreased (P 〈0.01). [Conclusion] Blood serum containing decoction of Four Drugs with Taohong could restrain migration of vascular smooth muscle ceils and its mechanisms are likely due to inhibition of the expression of intergrinβ3 and MMP9.
