摘要 目的:探讨生长分化因子-9(GDF-9)蛋白及其受体BMPR-ⅡmRNA在PCOS大鼠卵巢的表达。方法:皮下注射DHEA建立PCOS模型,用免疫组化法检测卵巢GDF-9表达;用RT-PCR法检测BMPR-ⅡmRNA的表达。结果:GDF-9主要于PCOS发育正常的初级、窦前、窦状卵泡卵母细胞胞浆中表达,表达强度与对照组的差异无统计学意义(P>0.05)。BMPR-ⅡmRNA相对含量则显著高于对照组(P<0.05)。结论:GDF-9是促进P- COS卵泡发育的主要原因;受体表达上调是诱导雄激素增高的原因之一。 Objective:To study the expression of GDF-9 protein and its receptor BMPR-Ⅱ mRNA in the PCOS rats. Methods:Established PCOS rat model by subcutaneous injecting DHEA. The expression of GDF-9 protein in PCOS groups, and control groups were detected and evaluated by iimnunohistochemistry and image analysis system. The transcription of GDF-9 receptor BMPR-Ⅱ mRNA, in PCOS groups and control groups, were measured and evaluated by semi-quantitative RT-PCR (reverse transcription polymerase chain reaction) and gel image analysis system. Results: GDF-9 protein expressed mainly in the ooeytes of primary, preantral and antral follicles. And there was no significant difference of expression intensity ( P 〉 0.05 ). Compared with the control groups, the gel image analysis system demonstrated that the transcription of BMPR- Ⅱ mRNA were increased significantly in PCOS groups ( P 〈 0.05 ). Conclusion: GDF-9 can play an important role in mainteinng follicles development, the increasing expression of GDF-9 receptors in PCOS enhance one of the function of GDF-9 in inducing androgen secretion.
