白细胞介素-8与子宫内膜异位关系

Effect of IL-8 on pathogenesis of endometriosis

目的探讨白细胞介素-8(IL-8)在子宫内膜异位症中(简称内异症)的表达及在发病机制中作用。方法将内异症患者按修订后的美国生殖协会子宫内膜异位症分期法(R-AFS)进行分期,分别用酶联免疫吸附法(ELISA)检测腹腔液中IL-8表达水平,实时荧光定量PCR检测子宫内膜组织IL-8 mRNA水平,以log互补DNA/log3-磷酸甘油醛脱氢酶(log cDNA/log GAPDH)代表其mRNA水平。结果内异症患者与对照组腹腔液中IL-8的表达水平分别为(23.37±8.23),(12.32±4.27)pg/ml,差异有统计学意义(P<0.05),内异症增生期和分泌期内膜组织IL-8 mRNA表达水平分别为1.3011±0.2689,1.1239±0.2738,2者差异无统计学意义(P>0.05)。结论IL-8作为自分泌生长因子和正反馈因子,直接促进内膜基质细胞生长,在内异症的发生和发展中起重要作用。

Objective To explore the expression of interleukin- 8 （IL- 8） and its effect on pathogenesis of endometriosis. Methods The patients were divided into different groups on base of R - AFS standard, and the levels of IL - 8 in peri- toneal fluid and IL - 8 mRNA in endomembrane tissue were detected by ELISA, real - time - PCR, respectively. The level of IL - 8 mRNA was represented by lg cDNA/lg GAPDH. Results The levels of IL - 8 in peritoneal fluid and endomembrane tissue were （23.37 ± 8.23 ） pg/ml, （ 12.32 ± 4.27 ） pg/ml, respectively （ P 〈 0.05 ）. However, the expression of IL - 8 mR- NA in estrin phase of endomembrane was 1. 3011 ± 0. 2689, 1.1239 ± 0. 2738（log cDNA/log GAPDH）. It was similar in two groups （P 〉 0.05）. Conclusion IL - 8, a kind of autocrine growth factor and positive feedback factor, can promote the growth of stroma cells and play a key role in pathogenesis of endometriosls.
