退变颈椎间盘组织中细胞凋亡及相关蛋白Caspase-8、Caspase-9的表达 被引量:4

摘要 目的探讨凋亡相关蛋白Caspase-8、Caspase-9在颈椎间盘退变中的作用。方法采用原位DNA片段末端标记和免疫组织化学方法,检测57例颈椎病退变颈椎间盘组织细胞及42例创伤对照颈椎间盘组织细胞凋亡状态及凋亡相关蛋白Caspase-8、Caspase-9的表达。结果57例颈椎病退变颈椎间盘组织细胞凋亡率(AI)为54.0%,Caspase-8蛋白阳性细胞百分率为43.0%,Caspase-9蛋白阳性细胞百分率为54.0%;42例创伤对照颈椎间盘组织细胞凋亡率(AI)为39.6%,Caspase-8蛋白阳性细胞百分比为35.4%,Caspase-9蛋白阳性细胞百分比为47.0%。两组间细胞凋亡率(AI)、Caspase-8、Caspase-9蛋白表达差异均有显著性意义(P<0.05)。凋亡细胞数越多,Cas-pase-8、Caspase-9表达越强。结论Caspase-8、Caspase-9蛋白参与了颈椎间盘组织中细胞调亡的调节,可能在颈椎病颈椎间盘退行性变发生和发展中发挥重要作用,且细胞调亡蛋白Caspase-9的作用可能比Caspase-8强。 Objective To explore the expression of apoptotic related protein Caspase -8 and Caspase -9 in degenerative cervical disc tissue. Methods We detected the apoptotic status and the expression of Caspase-8 and Caspase-9 in 57 cases of degenerative cervical intervertebal disc tissues and 42 of traumatic normal cervical intervertebal disc tissues with TdT-mediated dUTP-biotin nick end labeling (TUNEL) and immunohistochemistry methods. Results The average apoptotic index (AI) in 57 cases of degenerative cervical intervertehal disc was significantly higher than that of 42 traumatic normal cases (54.0% vs. 39.6%, P 〈0.01 ). The expressions of Caspase-8 and Caspase-9 protein were in the chondrocytes of cervical intervertebal disc tissues. The expression rates of Caspase-8 and Caspase-9 protein in 57 cases of degenerative cervical intervertebal disc were 43.0% and 54.0%, respectively. Which were significantly higher than those of 42 traumatic normal cases (35.4%, 47.0% ) ( P 〈 0.05), The stronger Caspase - 8 and Caspase -9 protein expressed, the bigger the apoptotic index was. Conclusion Both Caspase -8 and Caspase-9 may play an important role in the occurrence and progression of cervical intervertebal disc degeneration, and expression of the Caspase-8 protein is stronger than that of Caspase-9.
