激素性股骨头缺血性坏死局部微循环变化与桃红四物汤的干预作用 被引量:8

摘要 目的:研究证实桃红四物汤可对抗激素的作崩,对早期激素性股骨头缺血性坏死有防治作用。制备兔典掣激素忾股骨头缺血性坏死模型,拟进一步验证桃红四物汤对激素性股骨头缺血性坏死血液流变学和局部微血管密度的影响。方法:实验于2005-04/2006—10在福建中医学院中心实验室完成,动物实验方法符合动物伦理学要求。①实验材料及分组:选用健康成年新西兰大白兔60只,其中54只采用胡氏造模法制作激素性股骨头缺血性坏死模型,其余6只为正常组。造模6周后麻醉处死6只模型兔,与正常组对比确定造模成功。将42只造模成功兔按随机数字表法分为激素对照组和中药治疗组(n=21)。②实验方法及评估:激素对照组以生理盐水7mL/kg治疗,1次/d:中药治疗组以桃红四物汤7mL/kg治疗,1次/d。于治疗后第1,2,4周各组分别处死7只动物,检测血液流变学指标及股骨头局部微血管密度。结果:42只兔全部进入结果分析。①治疗后第1,2,4周激素对照组兔全血高切和低切黏度、血浆黏度、红细胞压积、纤维蛋白原含量均高于中药治疗组(P〈0.05~0.01),并且随着中药治疗时间的延长,中药治疗组咀液流变学趋于正常,与激素对照组的差异更明显。②治疗后第1,2,4周中药治疗组兔股骨头局部微血管密度均高了二激素对照组(P〈0.05-0.01)。结论:桃红四物汤能降低激素性股骨头缺血性坏死兔血液黏度,促进股骨头局部微血管的再生,改善股骨头局部血运,促进坏死股骨头的修复。 AIM: Some studies have proved Taohong Siwu decoction can resist hormone, and prevent glucocorticoid-induced avascular necrosis of the femoral head in early stage. In this study, we investigated the effect of Taohong Siwu decoction on hemorheology and microvessel density of rabbits with glucocorticoid-induced avascular necrosis of femoral head. METHODS: The experiment was performed at the central laboratory of Fujian College of Traditional Chinese Medicine from April 2005 to October 2006. All animal treatments were accorded with the ethical requirements. ①Fifty-four of sixty healthy adult New Zealand rabbits were made into models of glucocorticoid-induced avascular necrosis of femoral head using He's modeling method, and the other 6 rabbits served as normal. Six modeled rabbits were executed after 6 weeks, and identified the model were successful by comparing with normal group. Forty-two successfully modeled rabbits were randomly divided into treatment group and control group (n =21). ②The control group was administrated with normal saline 7 mL/kg daily, and the healing group was administrated with Taohong Siwu decoction 7 mL/kg daily. After 1, 2 and 4 weeks, 7 animals in each group were executed to detect hemorheological indexes and microvessel density of femoral head. RESULTS: Forty-two rabbits were all included in the final analysis, ①Whole blood viscosity at high and low shear rates, plasma viscosity, hematocrit, and fibrin content in control group were all significantly higher compared with those in treatment group in the 1^st, 2^nd, and 4^th weeks after treatment (P 〈 0.05-0.01). The hemorheology in treatment group recovered to normal state with the treatment time, and the differences between two groups were more significant.②Microvessel density of femoral head in treatment group was higher than that in control group in the 1^st, 2^nd, and 4^th weeks after treatment (P 〈 0.05-0.01 ). CONCLUSION: Taohong Siwu decoction can decrease blood microcirculation, and accelerate the repair of necrotic femoral femoral head. viscosity, promote the regeneration of microvessel, improve local head in rabbits with glucocorticoid-induced avascular necrosis of
