胸腺素β4促进创伤愈合过程中对多种生长因子及新生细胞迁移的作用 被引量:8

摘要 学术背景:近年来,胸腺素β4的促进创伤愈合作用受到关注。最近的研究发现,在心肌、角膜和皮肤的创伤愈合过程中,胸腺素β4主要是作为一种化学诱导因子,调节多种生长因子的表达以及趋化新生细胞的迁移。目的:阐述胸腺素β4的生物学性质及创伤愈合的机制,并重点就胸腺素β4促进皮肤创伤愈合的作用及机制进行分析。检索策略:文章作者应用计算机检索Medline1980-01/2007-08有关胸腺素β4促进创伤愈合机制的文章,检索词为"thymosinβ4,skin wound healing,myocardium wound healing,cornea wound healing",限定文章语言种类为English。同时检索中国期刊全文数据库2000-01/2007-08有关胸腺素β4促进创伤愈合机制的文章,检索词为"胸腺素,皮肤创伤愈合,心肌修复,角膜修复",并限定文章语言种类为中文。初检得到660篇文献,全部为机检。文献评价:阅读标题和摘要对660篇文献进行初筛,排除研究目的与本课题无关及内容重复性研究,保留218篇中英文文献进一步分析,其中55篇综述类文献,163篇研究论文。按纳入标准筛选,共126篇文章满足全部纳入标准,予以纳入。其中23篇文献研究了胸腺素β4的生物学特性,51篇研究了胸腺素β4与皮肤愈合的问题,33篇研究了胸腺素β4与心肌损伤修复的问题,19篇研究了胸腺素β4与角膜修复的问题。资料综合:由于机体自身修复愈合的能力极其有限,一旦发生损伤或者病变很难自愈,甚至导致溃疡或瘢痕形成,多需要外缘性修复或启动内源性修复,但目前尚未发现有效的促进创伤愈合的药物。近年来,胸腺素β4促进创伤愈合的作用受到关注,为创伤愈合药物的研究提供了一个新方向。结论:虽然有很多研究证实胸腺素β4在创伤愈合过程中有重要作用,但是具体作用机制尚未完全明了。目前,人工合成的胸腺素β4已用于大量实验,以探讨其在创伤愈合中的作用和机制,结果显示胸腺素β4很有可能成为促进创伤愈合,尤其是糖尿病溃疡、免疫功能低下慢性损伤的新的治疗药物。 BACKGROUND: In recent years, the promoting effect of thymosin β4 on wound healing has received great attention. Studies have found during the wound healing of myocardium, cornea and skin, thymosin β4 mainly acts as a kind of chemical inducing factor to regulate the expression of various growth factors and promote new cell migration. OBJECTIVE: To explain the biological characters of thymosin β4 and the mechanisms for wound healing, especially the mechanisms of thymosin β4 to promote skin wound healing. RETRIEVAL STRATEGY: A computer-based online search of Medline was conducted to identify the English literatures related to the mechanisms of thymosin β4 to promote wound healing published between January 1980 and August 2007 with the keywords of "thymosin 13 4, skin wound healing, myocardium wound healing, cornea wound healing". Meanwhile, we searched CNKI database for relevant Chinese articles dated between January 2000 and August 2007 with the same keywords in Chinese. 660 literatures were firstly retrieved by computer. LITERATURE EVALUATION: 660 articles were selected firstly by reading the titles and abstracts to exclude the articles unrelated with the subjects and repetitive studies, 218 articles were selected including 55 review articles and 163 study papers. According to the inclusive criteria, 126 articles were in accordance with the criteria. Of them, there were 23 about the biological characteristics of thymosin β4, 51 about thymosin β4 and wound healing, 33 about thymosin β4 and myocardial injury and repair, and 19 related to thymosin β4 and corneal repair. DATA SYNTHESIS: Due to the limited self-repair ability of body, once there is lesion or injury, it is hard to cure by itself, even ulcer or scar forms. Therefore, extrinsic factors or endogenic repair are indispensable. However, so far no effective drug has been found for wound healing. Recently, people have paid close attention to thymosin β4 for promoting wound healing, which designates a direction for the pharmaprojects on wound healing. CONCLUSION: A lot of researches have proved that thymosin β4 plays a key role in wound healing, but the mechanisms are not clear. At present, recombinant thymosin β4 has been used in lots of research to expiore its effect and mechanisms for wound healing. The results show that thymosin β4 may be the novel drug to promote wound healing, especially diabetic ulcers, chronic injury with immune function deficient.
