摘要
目的:探讨中药YN-3号对慢性应激模型大鼠抑郁状态相关行为学的影响及其作用机制。方法:40只SD大鼠分为空白组、模型组、YN-3组和氟西汀组,应用慢性复合应激方法造模,进行抑郁状态相关行为学检测,包括Open-field实验,强迫游泳实验和"Y"迷宫实验。免疫组化方法检测海马内GR、NMDAR表达情况。结果:与空白组比较,模型组大鼠的探究活动减少(P<0.01),强迫游泳持续时间延长(P<0.01),学习记忆能力降低(P<0.01),YN-3组大鼠相应行为学实验比模型组有显著提高(P<0.01,P<0.05,P<0.05);模型大鼠海马内GR、NMDAR阳性表达强度显著降低(P<0.01),YN-3组海马内GR、NMDAR的阳性表达强度有所提高(P<0.01)。结论:提示中药YN-3号可以减轻慢性应激引起大鼠抑郁状态的程度,其机制可能与脑内GR、NMDAR表达变化有关。
Objective :To investigated the influence of Chinese formula YN -3 on behaviors and GR, NMDAR in hippocampal of the chronic stress depression rats. Methods : Forty SD rats were divided into control, model, YN -3 and Prozac groups. Chronic stress model was established by using chronic mixed stimulation, Observe the behaviors caused by ehronic stress, including changes of spontaneous movements in Open -field test, learning- memory behavior in " Y" - maze test and stillness time in forced - swimming test. The expression of GR and NMDAR in hippoeampal tissue was detected by immunohistochemical technology. Results : Compare with control group, the results of spontaneous movements, stillness swimming time and learning - memory behaviors of the chronic stress model rats were declined significandy ( P 〈 0. 01, P 〈 0.01, P 〈 0.01 ), which of the Y N -3 group were improved compared with model group ( P 〈 0.01, P 〈 0.05, P 〈 0.05 ) ; the expression of GR and NMDAR in hippocampal of model rats was declined significandy compared with control group( P 〈0.01 ) and which was increased in the YN-3 group( P 〈0.05, P 〈0.01 ). Conclusion : The results show that YN - 3 could reduce the changes of behaviors which were caused by chronic stress, the possible mechanism of it might related to the changes of expression of GR and NMDAR in the hippocampi.
