超声激活血卟啉声动力作用对鸡胚绒毛尿囊膜VEGF表达的影响

摘要 目的:本研究探讨超声激活血卟啉(Hematoporphrin,Hp)声动力作用对鸡胚绒毛尿囊膜(Chorioallantoie membrane,CAM)的血管内皮细胞生长因子(Vascular endothelial cell growth factor,VEGF)表达的影响,为应用声动力疗法(Sonodynamic therapy,SDT)进行抗肿瘤血管新生提供实验基础。方法:种蛋随机分为4组,孵育7d后建CAM模型,随机分为空白组、单纯超声组、单纯Hp组、声动力治疗组。各组药物分别加于CAM表面的载体上,作用后制备标本,用免疫组化检测各组鸡胚CAM VEGF表达情况。结果:与空白组相比,单纯Hp组差异无统计学意义(P>0.05);单纯超声组、声动力治疗组与之差异有统计学意义(P<0.05);单纯超声组与声动力治疗组之间比较,差异有统计学意义(P<0.05)。结论:Hp介导的声动力作用显著抑制了鸡胚绒毛尿囊膜VEGF的表达,这可能是其抑制新生血管形成的重要机制。 Objective:To investigate the effect of sonodynamic therapy with hematoporphrin on the expression of VEGF in Chick Chorioallantoic Membrane and provide experimental basis for appling sonodynamic therapy(SDT) on anti-angiogenesis in tumor therapy. Methods:The hatching eggs cultured for 7 days were randomly divided into 4 groups: blank control group,ultrasound group,hematoporphrin group,sonodynamic therapy group. The express of VEGF in CAM was detected by immunity histochemistry. Result:There were significant differences between control group and ultrasound group(P〈0.05) and also between control group and SDT group(P〈0.05),and between ultrasound group and SDT group(P〈0.05);while no difference between control group and hematoporphrin group(P〉0.05), conclusion The results show that sonodynamic therapy with hematoporphrin has a significant inhibitionary effect on the express of VEGF in CAM and this maybe is the most important mechanism of its anti-angiogenesis effect.
