幽门螺杆菌致BALB/c小鼠胃粘膜内肥大细胞及其脱颗粒变化的研究 被引量:4

摘要 目的:探讨幽门螺杆菌(Helicobacter pylori,Hp)对小鼠胃粘膜内肥大细胞(Mast cells,MC)及其脱颗粒的影响,从而证明Hp在慢性荨麻疹(Chronic urticaria,CU)的发病过程中的重要作用。方法:50只健康BALB/c小鼠随机分成2组,Hp组30只,每间隔3d灌喂1次Hp菌液,共7次,牛血清白蛋白(Bovine albnmin seFum,BAS)为对照组,灌胃2%的BAS,灌喂方式同上。用甲苯胺蓝染色法测定灌胃后老鼠胃粘膜内MC总数及脱颗粒的百分比,每片连续计数10个高倍视野的MC,结果以个数/高倍视野表示,脱颗粒MC比值以脱颗粒MC总数/MC总数表示,并与对照组比较。结果:2组小鼠胃粘膜MC总数间的差异有统计学意义(100.0比30.7,P<0.01);脱颗粒MC总数间的差异有统计学意义(48.85比6.51.P<0.01);脱颗粒MC百分比之间的差别有统计学意义(50%比23%,P<0.01)。结论:Hp感染可导致小鼠胃粘膜MC总数增加,并且脱颗粒MC所占的比例亦显著增加.从而释放组胺和一系列血管活性介质到外周血,这可能与导致临床上的红斑。 Objective:To search the impact of helicobacter pylori infection in changes of mice gastric mucosa mast cells and degranulation process,therefore to establish the important role of Hp in the patbogenesis of chronic urticaria. Methods:50 BALB/c healthy mice were randomized into two groups:Hp group,consist of 30 mice,were given Hp liquid every 3 days for total 7 times. Bovine albumin serum(BAS) as control group,was given BAS 2% with the same way as Hp group. Toluidine blue staining of the gastric mucosa was used to assess the total number of mast cells,the amount of degranulated mast cells and the percentage of degranulation after feeding. Each slide was consecutive count with 10 high-power fields for mast cells; resuhs express with number of mast cells/ high power field, the ratio of degranulated mast cell was expressed by the amount of degranulated mast cells/ total of mast cells. At the same time compared with the control group. Results:There was significant statistical difference in the total number of mast cells between the tow groups(100.0vs0.7,P〈0.01);total degranulated mass cells among two groups bad a statistic significant different(48.85vs6.51 ,P〈0.01 );degranulated mast cell percentage among two groups had a statistic significant different(50%vs23%,P〈0.01). Conclusion:Hp infection increases the number of gastric mucosa' mast cells,also significantly increases degranulated mass cells proportion,thus release histamine and a series of vascular activation transmitter to peripheral circulation that may cause a vascular mucosa dilation, wheal,etc. A typical urticaria sign.
