摘要
目的:探讨增殖细胞核抗原(PCNA)和第10号染色体缺失性磷酸酶和张力蛋白同源物基因(PTEN)在脑胶质瘤组织中表达差异的临床意义.方法:采用组织芯片技术建立96例脑胶质瘤和22例正常脑组织的组织芯片,并用免疫组织化学Elivision法,分别检测PTEN和PCNA在不同组织学类型和不同组织学分级的人脑胶质瘤组织和正常脑组织中的表达差异.结果:PCNA和PTEN在脑胶质瘤组织中的表达与在正常脑组织中的表达之间的差别有显著意义;PTEN在星形细胞瘤中的表达阳性率45/78(57.7%)显著高于其在非星形细胞瘤中的表达阳性率5/18(27.8%)(P〈0.05);PCNA在高分化脑胶质瘤(组织学分级Ⅰ-Ⅱ级)中表达低于在低分化脑胶质瘤(组织学分级Ⅲ-Ⅳ级)中表达;随着胶质瘤恶性度增高,PTEN表达强度逐渐下降,PCNA的表达则呈上升趋势.结论:PCNA的表达反应了脑胶质瘤细胞增殖与分化的一般规律,PTEN表达的降低在脑胶质瘤的发生、发展过程中可能起着非常重要的作用,PCNA和PTEN的联合检测有利于更准确地判断胶质瘤生物行为和评估患者预后.
AIM: To investigate the clinical significance of the differential expressions of PCNA and PTEN in glioma tissue and normal brain tissue. METHODS : Tissue microarray including 96 gliomas and 22 normal brain tissues was prepared. The expres- sions of PCNA and PTEN in glioma tissue and normal brain tissue with different histological types and different histological grades were detected by Elivision method of immunohistochemistry. RESULTS : The positive expression rates of PCNA and PTEN had signifieandy difference between glioma tissue and normal brain tissue ( P 〈 0.05 ). The expression of PTEN was significantly higher in astrocytoma than in other types of glioma ( P 〈 0.05 ) ; the expression of PCNA was significantly lower in well-differen- tiated glioma than in poorly differentiated glioma, but the expression of PTEN was significantly higher in well-differentiated glioma than in poorly differentiated glioma; along with the malignant level of glioma rising up, the expression intensity of PTEN decreased, and the expression of PCNA increased. CONCLUSION: The expression of PCNA mirrors the general rule of cell multiplication and differentiation of glioma. The decrease of the expression of PTEN may have very important effect on development and growth of glioma. Combined detection of PCNA and PTEN can estimate the biological behaviour and prognosis of the patients more exactly.
