Expressions of hypoxia-inducible factor and vascular endothelial growth factor in clear cell renal cell carcinoma

摘要 目的:研究肾透明细胞癌(CCRCC)中HIF-1α、HIF-2α的表达与VEGF、MVD的相关性及其对肿瘤病理分期的影响。方法:在40例CCRCC组织标本中,应用免疫组化方法检测HIF-1α、HIF-2α和VEGF的表达,用CD34单克隆抗体标记肿瘤微血管密度(MVD)。结果:HIF-1α染色阳性28例,HIF-2α染色阳性25例,VEGF染色阳性24例。Spearman等级相关分析发现HIF-1α、HIF-2α分别与VEGF表达程度呈正相关。HIF-1α、HIF-2α或VEGF阳性肿瘤组织的MVD值显著高于正常肾组织(P<0.05)。未发现HIF-1α、HIF-2α和VEGF表达与肿瘤病理分期有关(P>0.05)。结论:在CCRCC中HIF-1α、HIF-2α与VEGF表达相关,并可能诱导VEGF的过度表达,促进肿瘤血管生成,而且与CCRCC的侵袭性生物学行为密切相关,但其表达与肿瘤病理分期无相关性。 Objective:To investigate the expressions of hypoxia-inducible factor (HIF-1α, HIF-2α) and vascular endothelial growth factor (VEGF) in clear cell renal cell carcinoma (CCRCC) and their correlations with microvascular density (MVD) and tumor staging. Methods: Expressions of HIF-1α, HIF-2α and VEGF were detected by immunohistochemistry in 40 cases of CCRCC. MVD was marked by monoclonal antibodies CD34 markers. Results: HIF-1α and HIF-2α expressions were positive in 28, 25 cases, and VEGF expression was positive in 24 cases. Spearman correlation analysis showed the positive correlations between HIF-1α, HIF-2α and VEGF respectively. MVD in positive tissues was significantly higher than that in negative ones (P〈0.05). There was no relation between the HIF and VEGF expressions and tumor staging (P〉0.05). Conclusion:The expressions between HIF-1α, HIF-2α and VEGF are positive in most CCRCC tissues, which could induce over-expression of VEGF and promote tumor angiogenesis and invasiveness. But there is no relationship between HIF-1α, HIF-2α and VEGF expressions and tumor staging.
