Replacement of primary and permanent teeth contains a series of complicated procedures, in which root and alveolar bone resorption is an essential part. In the past, the researches in RANKL/RANK pathway mostly focus on its regulation in osteoclast. It is believed that RANK brings the cellular signals into pre-osteoclasts, in which they induce osteoclasts to generate, differentiate, and activate bone resorption. However, RANKL/RANK pathway and its regulation in root and alveolar resorption, especially in odontoclast, attract more and more scientists' interests recently. This review introduces the recent research in RANKL/RANK pathway and its regulation in root and alveolar resorption. Plenty of researches indicate that in root and alveolar resorption, RANKL/RANK pathway may play a similar role in osteoclasts as that in odontoclasts. In addition, varied cytokines possibly involved ha this procedure are also introduced in this review, and they may positively or negatively regulate the root and alveolar resorption.
