摘要
目的研究非结构蛋白5A反式激活蛋白6(Ns5ATP6)基因在细胞内的表达及表达蛋白的亚细胞定位。方法构建HBeAgTP基因的绿色荧光蛋白表达质粒pEGFP-Cl-NS5ATP6,pEGFP-Cl-NSSATP6转染HepG2细胞,24h后荧光显微镜下观察蛋白表达的亚细胞定位。结果成功构建出NS5ATP6基因绿色荧光蛋白表达质粒pEGFP-Cl-NS5ATP6,其表达的蛋白亚细胞定位于细胞质。结论NS5ATP6基因可表达蛋白,其表达蛋白亚细胞定位于细胞质。
Objective To found the subcellular location of the human gene 6 transactivated by nonstructural protein 5A of hepatitis C virus (NSSATP6). Methods Green fluorescent protein (GFP) expression vector pEGFP- NS5ATP6 was established. The pEGFP- NS5ATP6 was transfected into HepG2 cells, and analyze the subcellular location of the proteins expressed by NSSATP6 through Green fluorescent microscopy after 24 hours. Results The pEGFP-NSSATP6 gene was successful cloned, NSSATP6 can express protein in cells and subcellularly located in cell plasma. Conclusion NSSATP6 can express protein, and the protein expressed by NSSATP6 subcellularly located in cell plasma.
