摘要
目的:探讨缺氧诱导因子-1α(HIF-1α)和葡萄糖转运蛋白-1(GLUT-1)在膀胱移行细胞癌中的表达及与膀胱移行细胞癌生物学行为的关系。方法:应用免疫组化方法对50例膀胱移行细胞癌及正常膀胱组织中HIF-1α和GLUT-1的表达进行检测,并用计算机图象分析系统对其表达水平进行定量分析。结果:正常膀胱组织中HIF-1α和GLUT-1均不表达,50例膀胱移行细胞癌中,HIF-1α和GLUT-1的阳性表达率分别为52%和76%。HIF-1α的表达水平与膀胱移行细胞癌的分级和复发密切相关(P<0.05),但与分期无关;GLUT-1的表达水平与膀胱移行细胞癌的分级、分期和复发均密切相关(P<0.05)。HIF-1α和GLUT-1在膀胱移行细胞癌中的表达水平亦呈正相关(r=0.381,P=0.029)。结论:膀胱移行细胞癌中HIF-1α和GLUT-1的表达与肿瘤的恶性进展密切相关,联合检测HIF-1α和GLUT-1可作为了解膀胱移行细胞癌生物学行为的参考指标,并有可能为膀胱移行细胞癌的基因治疗提供新的靶点和新的思路。
Objective: To investigate the relationship between the expression of hypoxia-inducible factor-1α (HIF-1α) and glucose transporter-1 (GLUT-1) and the biological behavior of bladder transitional cell carcinoma. Methods:The expression of HIF-1α and GLUT-1 in 50 cases of bladder transitional cell carcinoma was detected by immuno- histochemical method and the intensity of HIF-1α and GLUT-1 expression was assessed by computer image analysis system. Results: HIF-1α and GLUT-1 were not expressed in normal bladder tissue. But in bladder transitional cell carcinoma, the rates of HIF-1α and GLUT-1 expression were 52 M and 76 M respectively. The expression of HIF-1α was positively associated with the grade and recurrence (P〈0.05), but its expression was not correlated with staging. The expression of GLUT-1 was also positively associated with the grade, staging and recurrence (P〈0. 05). And the expression of HIF-1α had a positive correlation with GLUT-1 (r= 0. 381, P= 0. 029). Conclnsions:The expression of HIF-1α and GLUT-1 is strongly associated with neoplastic progression in bladder transitional cell carcinoma. United detection of HIF-1α and GLUT-1 may be one of parameters for understanding the biological behavior of bladder transitional cell carcinoma. And further study of HIF-1α and GLUT-1 may provide a new target and idea for the gene therapy of bladder transitional cell carcinoma.
