针刀松解法对第3腰椎横突综合征模型大鼠耐痛阈及脊髓与下丘脑ENK、β-EP的影响 被引量:18

Effect of acupotomy dissolution on pain threshold and ENK and β-EP in spinal cord and hypothalamus in rats with transverse syndrome of the third lumbar vertebrae

摘要 目的观察大鼠痛阈及下丘脑与脊髓脑啡肽(enkephalin,ENK)、β内啡肽(β-enkorphin,β-EP)的含量变化,探讨针刀松解法治疗第3腰椎横突综合征的中枢镇痛机制。方法SD大鼠,随机分为4组:正常组、模型组、电针组、针刀组,后3组首先建立第3腰椎横突综合征的动物模型,后2组造模后分别给予电针和针刀松解干预治疗。分别在9个时间点测量各组大鼠痛阈,存活28d后取大鼠下丘脑和脊髓腰段,以酶联免疫方法检测大鼠下丘脑与脊髓腰段中ENK、β-EP的含量。结果模型组大鼠痛阈降低,脊髓与下丘脑中ENK、β-EP的含量比正常组明显升高(P<0.01);针刀松解法和电针治疗组大鼠痛阈提高,脊髓与下丘脑中ENK、β-EP的含量比模型组明显降低(P<0.01),接近正常组水平,其中针刀组的含量较电针组更接近正常组(P<0.05)。结论针刀松解法与电针能提高第3腰椎横突综合征模型大鼠痛阈,并可良性调节中枢神经系统内ENK、β-EP的合成和释放,使其接近正常水平,针刀松解法的镇痛效果略优于电针治疗。 Objective To observe the changes of pain threshold and ENK and β-EP contents in the spinal cord and the hypothalamus, and to study the central analgesic effect of acupotomy dissolution in rats with transverse syndrome of the third lumbar vertebrae. Methods Twenty-eight SD rats were randomly divided into normal control group, model control group, acupotomy dissolution group and electroacupunture( EA )group, with 7 eases in each group. Model was established by operation. Acupotomy dissolution was applied twice, EA was applied six times. Radiation heat stimulation was applied to the rat tail and the pain threshold(PT) was detected by tail flick latency test. The test would be done 9 times. ENK and β-EP contents in spinal cord and the hypothalamus were detected with enzyme linked immunosorbent assay(ELISA) on day 28. Results In the model group, the pain threshold values were lower than in the normal group and ENK and β-EP contents in the spinal cord and the hypothalamus were increased significantly (P 〈0.01 ). In the acupotomy dissolution group and EA group, the pain threshold values were higher than in the model control group, and ENK and β-EP contents in the spinal cord and the hypothalamus were decreased significantly ( P 〈 0.01 ). ENK and β-EP contents in the acupotomy dissolution group were closer to those in the normal control group than in the EA group (P 〈 0.05 ). Conclution Acupotomy dissolution and EA can raise the pain threshold value by adjusting ENK and β-EP of central nervous system. Acupotomy dissolution was more effective.
