摘要
采用C-70培养基培养钩端螺旋体可提高钩体浓度4.5~11.6倍。参照Auran方法进行提纯,采用中空纤维超滤技术收集外膜制成外膜苗苗。接种一针(30μg)即可达到最低免疫水平,表明外膜苗有较强的免疫原性。尽管疫苗中残留微量SDS,但动物试验安全,未出现异常反应。其它检定项目结果均符合构体菌苗制检现程要求。
By using the C-70medium prepared by the authors, the culture concentration of leptospira was increased by 4. 5~ 11. 6folds. After the cultured leptospira was purified by hollow fibre ultrafiltration as described by Auran, the outer membrane of it was collected for preparing vaccine. The outer membrane protein vaccine showed good immunogenicity. The antibody of animals immunized with only a single does (30μg) of the outer mernbrane vaccine reached the minimal protective level. Though the vaccine contained a little residual SDS, no adverse reaction was found during safety test. All the results of other control testings met the current national requirments of leptospira vaccine.
