摘要
应用细胞原位杂交技术,观察经重组小鼠白细胞介素-19(IL-1β)处理后的体外培养的新生1d大鼠中脑黑质神经元c-jun基因的表达.结果显示,培养的黑质细胞多为酪氨酸羟化酶阳性神经元,IL-1β可诱导体外培养的黑质神经元c-junmRNA表达,高水平的表达出现在IL-1β处理后2~4h。说明IL-1β有兴奋黑质神经元的作用,并提示黑质神经元上可能存在IL-1β受体.
Interleukin-lb(IL-1β) was synthesized and reIeased by certain brain cell8 including neurons, atrocytes, brain macrophages and brain vessel endothelial ce1ls. IL-1 recept0rs were distributed in various brain regions. IL-1 was believed to play important role as a pleiotropic cytqkine in central nervous system. c-jun expression in neuronal network could be stimulated by various factors and was used as a signal of neuronaI excitement. In attempt to understand the possible action of ILl5 on mesencephalic substantia nigra neuron, c-jun tnRNA expression was examined in cultured substantia nigra neurons ' from l day postnataI rat midbrain after IL-1g(mouse recombinated IL-10)treatrnent by rnean of cellular in situ hybridization technique. Results showed that most cultured neurons were tyro8ine hydroxylase-positive and treatfuent with IL-1 caused arapid and transient c-jun mRNA expression in neurons. High level expression of c-jun gene occurred 2~4 hour after treatment. These results suggested that IL-l5 Was able to excite the cu1tured substantia nigra neurons and IL-18 receptor probably existed in sub8tantia nigra.
