摘要
目的确定双调蛋白在人体睾丸组织和精子的表达特征,比较双调蛋白在正常人和隐睾症患者睾丸组织的表达差异。方法收集正常睾丸组织、精液精子和隐睾症患者的睾丸组织,采用反转录-聚合酶链反应(RT-PCR)和免疫组织化学的方法检测双调蛋白的表达水平。结果双调蛋白在正常睾丸组织和精子均有表达,主要分布于睾丸长型精子细胞和成熟精子尾部;与正常睾丸组织比较,双调蛋白在隐睾组织的表达显著减少、隐睾组织生精小管的表达基本消失。结论在隐睾症睾丸中双调蛋白的表达显著减少,提示双调蛋白表达与睾丸发育有一定关系。
Objectives: To identify the expression of amphiregulin in human testis and ejaculated spermatozoa and to compare the amphiregulin expression in the testis from healthy men and the patients with cryptorchidism.
Methods. Normal testicular tissues were collected from 3 healthy men dead by accident. The testicular tissues of cryptorchidists and semen samples were obtained from the patients in Peking University Shenzhen Hospital. RT-PCR and immunocytochemistry were used to detect the expression of amphiregulin in testicular tissues and ejaculated spermatozoa.
Results: The mRNA expression of amphiregulin was detected by RT-PCR in the testicular tissues from both healthy men and patients. Compared with that in healthy men, the expression of amphiregulin mRNA was significantly decreased in the patients with cryptorchidism. With immunocytochemistry, amphiregulin was mainly located on the tails of elongated spermatids and ejaculated spermatozoa, and the expression of amphiregulin in the cryptorchidists was significantly lower than that in healthy men, which is similar to the results from RT-PCR.
Conclusion: The expression of amphiregulin in the testis was significantly decreased in the patients with cryptorchidism, which suggested that amphiregulin expression was related to the development of testis.
