摘要
用免疫细胞化学方法结合体视学分析,研究了Wistar大鼠的听源性惊厥易感大鼠(P77PMC)惊厥和点燃后海马结构内生长相关蛋白表达的差异、结果显示:(1)P77PMC大鼠一次惊厥后,海马结构内有广泛的生长相关蛋白免疫反应产物沉积,其分布呈板层样;(2)P77PMC大鼠点燃后,生长相关蛋白免疫反应产物的分布特征与一次惊厥后相比较,无明显变化,但是,海马CA3区苔藓纤维层生长相关蛋白免疫反应产物的光密度值显著增加(P<0.01)。结果表明,听源性惊厥点燃能够诱导海马结构内生长相关蛋白表达增加。本文还对生长相关蛋白表达增加的生物学意义进行了讨论.
Audiogenic kindling is used to make an animal model of epilepsy in audiogenic seizure prone rats. The growth as-sociated protein (GAP-43) is a neuron-specific phosphoprotein that is highly expresscd during the deveIopment of the centralnervous system, nerve regeneration and the modulation of synaptic function. The present study investigated the difference ofGAP-43 expression in the hippocampal formation after single (AS) and kindled (KAS) audiogenic seizures in P77PMC rats,a strain of Wistar audiogenic seizure prone rats. Using immunocytochemistry and morphometry, we obtained the followingresults: (1 ) in AS rats, GAP-43-like immunostaining in the hippocampal formation was characterized by a laminar distribution pattern, and this pattern was similar to that observed in normal rats; (2) in KAS rats, the pattern of GAP-43 immunostaining in the hippocarnpal forrnation was approximately similar to that in AS rats. As compared with AS rats, the opticaldensity of GAP-43 immunostaining significantly increased in mossy fiber layer in CA3 (P<0. 01). These results indicatedthat audiogenic kindling could increaSe the expression of GAP-43, and there was a close relationship between GAP-43 ex-pression and sprouting of granule cell.
