摘要
目的:建立HPLC法测定乌梢蛇药材中尿嘧啶、黄嘌呤、次黄嘌呤的含量。方法:采用高效液相色谱法。色谱条件为DoamonsilC18(5μm,200mm×4.6mm)柱,以0.05mol/L磷酸水溶液(pH=8.4)为流动相;检测波长为254nm;流速:1mL/min。结果:尿嘧啶、黄嘌呤、次黄嘌呤分别在0.06-0.14μg、0.17-0.50μg、0.27-0.6μg范围内具有良好线性关系(r分别为0.9996、0.9998、0.9996);平均加样回收率分别为102.0%、98.4%、100.3%;RSD分别为2.10%、2.67%、2.89%。结论:该法可用于测定乌梢蛇药材中尿嘧啶、黄嘌呤、次黄嘌呤的含量,从而控制乌梢蛇药材的质量。
Objective :To establish the HPLC method to determine the content of uracil, xanthine, hypoxanthine in Zaocys. Methods :The HPLC analysis was carried out on DoamonsilODS- C18 column (5μm 200 × 4.6mm), 0.05mol/L (NH4 )2 HPO4 was used as the mobile phase. The detection wavelength was at 254nm, the flow rate was 1mL/min. Results :The linear range of uracil xanthine and hypoxanthine was 0.06 - 0. 14μg,0.17 - 0. 50μg,0. 27 - 0.6μg which indicated the linearity of this method is all right. , and the r were 0. 9996 0. 9998 0. 9996. The average recovery of them were 102.0% ,98.4% , 100.3% , the RSD were 2.10% ,2.67% ,2.89%. Conclution :This method can be applied in quantitative determination of nucleoside in Zaocys,and also control the quality of Zaocys.
