隔核毁损对甲基苯丙胺模型大鼠行为和学习记忆能力的影响 被引量:1

摘要 目的观察隔核毁损对甲基苯丙胺(MAP)模型大鼠行为、学习记忆能力的影响。方法先将90只SD大鼠单纯随机分为3组:对照组,MAP组,MAP+隔核毁损组,每组30只。大鼠腹腔内注射MAP制造精神分裂症动物模型,采用立体定位仪电极毁损隔核,进行刻板行为评分。对照组及MAP+隔核毁损组进入水迷宫实验;另60只SD大鼠在游泳训练后也进行水迷宫实验,随机分为训练组(无毁损)及训练组(毁损后),每组30只。通过实验检测大鼠的空间学习记忆能力。结果与MAP组比较,MAP+隔核毁损组大鼠刻板行为评分显著下降。MAP+隔核毁损组大鼠社会行为良好。水迷宫实验中,MAP+隔核毁损组在初期空间记忆能力存在缺陷。结论隔核毁损能够实现对大鼠精神分裂症阳性症状的改善,对大鼠初期学习记忆能力及空间定位能力有影响,对已获得的记忆影响不大。 Objective To observe the effects of septal nucleus lesioning on behaviors and learning and memory abilities in methamphetaminetreated (MAP) rats. Methods One hundred and fifty Sprague-Dawley rats were enrolled in this study. The first 90 rats were randomly designed into 3 groups: control group, MAP group, MAP + septal nucleus lesioning group (30 each). Methamphetamine (MAP) was injected intraperitoneaUy for establishing the schizophrenic rat model. The septal nucleus was destructed by direct current using stereotaetic technique. The stereotyped behavior scores were recorded in MAP-induced schizophrenic rats. The control group and MAP + septal nucleus lesioning group were observed in the Morris water maze. Then, the other 60 rats were randomly divided into 2 groups after swimming education: septal nucleus lesioning and non-lesioning groups (30 each), and both received water maze tests. The spatial learning and memory abilities were observed through the Morris water maze tests. Results Compared with the MAP group, the stereotyped behavior scores were significant reduced in MAP + septal nucleus lesioning group. In the MAP + septal nucleus lesioning group, the social behaviors were normal, and the water maze tests revealed defect in spatial memory ability in the early days. Conclusion Lesioning of the septal nucleus could improve some symptoms of schizophrenia, but influence the learning and memory and spatial orientation ability in the early days after the lesioning, with no impact on previously acquired memory in rats.
