黄酮类化合物柚皮苷抑制HaCaT细胞RANTES产生的研究 被引量:4

摘要 目的:分析肿瘤坏死因子α(TNF-α)和γ干扰素(IFN-γ)是否诱导体外培养角质形成细胞株HaCaT产生趋化因子RANTES,了解柚皮苷是否影响TNF-α和IFN-γ对合成RANTES的诱导作用。方法:TNF-α和IFN-γ刺激培养的HaCaT细胞,应用定量RT-PCR方法测定RANTES的mRNA表达,应用ELISA方法测定上清液中RANTES。结果:正常培养HaCaT细胞RANTES微弱表达,TNF-α和IFN-γ刺激使HaCaT细胞RANTES的mRNA表达与分泌量显著增加。柚皮苷0.5,0.25mmol.L-1能抑制TNF-α和IFN-γ诱导的RANTES分泌。结论:柚皮苷可抑制TNF-α和IFN-γ引起的HaCaT细胞分泌RANTES。 Objective: To determine the inhibition of naringin on tumor necrosis factor-α (TNF)-α and interferon garnma-γ(IFN- γ)induced-RANTES in cultured human keratinocyte line HaCaT. Methods. Cultured HACAT cells were treated with TNF-α and TFN-γ. With prelncubatlon of naringin, expression of mRNA of RANTES was analyzed by RT-PCR. Protein of RANTFS in the supernatants was determined by ELISA. Results: In normal cultured HaCaT cell, RANTES mRNA and proteins were detected, TNF-α and TFN-γ significantly enhanced the mRNA expression and protein release of RANTES. Naringin(0. 5, 0. 25 mmol· L^-1 ) inhibitsed TNF-α and TFN-α induced production of RANTES. Conclusion. Naringin can inhibit TNF-α and TFN-α induced production of RANTES.
