山羊痘直接荧光抗体检测方法的建立与应用 被引量:1

摘要 用兔抗山羊痘病毒IgG制备荧光抗体,检测了临床发病山羊皮肤痘疹切片和山羊痘病毒B、Y、LD和QL株感染的BHK-21细胞中的病毒抗原。结果,皮肤痘疹切片、感染细胞抹片及飞片均呈阳性,而正常山羊皮肤和细胞对照为阴性;荧光抗体的最适工作浓度为1∶32,且这种荧光可被山羊痘标准阳性血清特异性抑制。表明,建立的山羊痘直接荧光抗体检测方法能对临床发病山羊皮肤痘疹和感染细胞中的山羊痘病毒抗原进行定位检测,为临床诊断山羊痘提供了一种简单快速的方法。 The fluorescent antibody was prepared with IgG purified from rabbit anti-goatpox virus (GPV) serum and used to detect the antigen of GPV in pock samples and BHK-21 cells infected respectively with GPV strains B,Y,LD and QL. All the samples of the pock sections and the cell slides and smears of the infected cells were positive, but the control samples of normal skins and cells were negative, and the optimal working concentration of the fluorescent antibody was 1 : 32,and the fluorescence could be specifically inhibited by GPV standard positive serum. These results indicated that the antigen of GPV in pock sampies and cells-infected with GPV could be detected locationally by the developed fluorescent antibody test (FAT) ,and FAT was a simple and clipping method for clinical diagnosis of GPV.
