摘要
目的研究细胞周期蛋白CyclinD1和p27在非小细胞肺癌(NSCLC)组织中的表达及其临床意义。方法应用S-P免疫组化法检测63例非小细胞肺癌组织中CyclinD1和p27的表达,取20例肺部正常组织做对照,并结合临床资料进行分析。结果在非小细胞肺癌组织中CycinD1呈高表达,p27呈低表达,与对照组均有组间差异(P<0.05)。CyclinD1和p27的表达均与肿瘤的分化程度、淋巴结转移、临床分期有关(P<0.05),而与年龄、性别、组织学类型无关(P>0.05);NSCLC组织中CyclinD1与p27表达呈显著负相关(P<0.01)。结论CyclinD1癌基因和p27抑癌基因突变协同作用使非小细胞肺癌发生与发展。二者的检测可作为肺癌的早期诊断指标、评价非小细胞肺癌恶性程度和判断预后的重要指标。
Objective To investigate the expressions of CyclinD1 and p27 proteins in non-small cell lung cancer,and discuss the relationship between their expressions and tumor' s clinical and pathological characters. Methods The expressions of CyclinD1 and p27 proteins were detected by immunohistochemical staining(S-P) in 63 cases with primary NSCLC and 20 cases of normal lung tissues in control group, and the clinical data were analyzed. Results The high expressions of CyclinD1 and the low expressions of p27 were observed in non-small cell lung cancer. Statistical significance existed between the tumor group and the control group(P 〈 0.05). In the non-small cell lung cancer group, the aberrant expressions of CyclinD1 and p27 were correlated with the histologic differentiation, clinical stage and lympho node metastasis(P 〈 0.05). There was no relationship between CyclinD1 ,p27 expressions and age, sex or histological subtype (P 〈 0.05 ) ; expression of CyclinD1 was negatively correlated with expression of p27 (P 〈 0.01 ). Conclusions The mutations of oncogene CyclinD1 and suppression gene p27 may contribute to the occurrence and progression of non-small cell lung cancer. The detection of the proteins of CylinDl and p27 could be an important marker to evaluate the malignancy degree and the prognosis of non-small cell lung cancer.
