摘要
目的评价重组38kD、Ag85B和16kD蛋白3种重组蛋白的诊断价值。方法采用蛋白免疫印迹方法观察本实验室克隆表达的结核分枝杆菌38kD、Ag85B和16kD蛋白抗原用于结核病血清学诊断的灵敏度,及3种抗原联合应用诊断的灵敏度。结果共对62例结核病人血清和19例健康人血清进行了IgG抗体检测,结果显示重组38kD检测灵敏度为51.6%,假阳性率为5.2%;重组Ag85B灵敏度为35.5%,重组16kD的灵敏度22.6%,无1例健康人血清与重组Ag85B及16kD蛋白反应。3种蛋白联合诊断的灵敏度为77.4%,假阳性率为5.2%。结论多种蛋白抗原联合应用将是结核病血清学诊断试剂的发展方向。
In the present study, the diagnostic values of three Mycobacterium tuberculosis (Mtb)-specific proteins 38 kD, Ag85B and 16 kD were evaluated using the protein immunoblotting, in which the sensitive of testing in sera of 62 patient with tuberculosis for 38 kD, Ag85B, 16 kD and 38 kD/Ag85B/16 kD recombinant proteins were 51.6%, 35.5% , 22.6% and 77. 4% respectively. In 19 healthy individuals tested, no false positive result could be detect except for 38 kD and 38 kD/Ag85B/ 16 kD recombinant proteins; (the positive results in these two ps were both to be 5.2%). It is evident that combined use of these 3 recombinant proteins in the immunoblotting assay may be better for diagnosis , rather than the use of a single protein.
