摘要 目的探讨哮喘大鼠气道重塑的机制和细胞周期蛋白D1(Cyclin D1)、细胞周期素依赖性激酶4(CDK4)在哮喘气道重塑中的表达以及盐酸氨嗅索的干预效果。方法应用卵蛋白激发大鼠建立哮喘气道重塑模型。60只SD大鼠分为3组:正常组,哮喘组和干预组,每组20只。采用免疫组化技术和计算机图象分析系统对大鼠气道Cyclin D1、CDK4的表达和气道重塑进行研究。结果哮喘纽气道壁均可见明显炎症细胞浸润,气道壁厚度增加,Cyclin D1、CDK4阳性表达增加,与正常组和干预组比较差异具有统计学意义(P〈0.05)。结论Cyclin D1、CDK4参与哮喘气道重塑的发生,盐酸氨嗅索可减少气道壁炎症细胞浸润,预防气道重塑。 Objective To approach the mechanism of the airway remodeling in the asthmatic rats, the expression of Cyclin D1 and CDK4 in asthmatic rats with airway remodeling and the effect of the ambroxol' s intervention. Methods The model of asthmatic rats with airway remodeling was induced with OVA. 60 SD rats were divided into 3 groups randomly: normal group, asthmatic group and intervention group. Each group had 20 rats. The method of immunohistochemistry and techniques of computer - assisted image analysis were used to detect the changes of the airway remodelling and the expression of Cyclin D1 and CDK4. Results In the asthmatic group,the infiltration of inflammatory cells, thicknesses of airway walls and expression of Cyclin D1 and CDK4 were significantly increased compared with those of the other two groups ( P 〈 0.05 ). Conclusion Cyclin D1 and CDK4 take part in the development of the asthmatic airway remodeling. Ambroxol can reduce the infiltration of inflammatory cells around the airway walls, and prevent the develop ment of airway remodeling.
