摘要
目的观察细胞膜galectin-1抗体阻断对黏附LST-R1细胞凋亡的影响。方法激光共聚焦显微镜检测galectin-1在LST-R1细胞的表达。galectin-1抗体预孵LST-R1细胞后种植于Ⅰ型胶原(20μL/孔)包被的48孔板中,流式细胞仪检测黏附LST-R1细胞凋亡变化。结果galectin-1表达于LST-R1细胞膜表面,galectin-1抗体预孵LST-R1细胞后结合率为(67.2±6.73)%。galectin-1抗体阻断组黏附细胞呈不规则角形,而对照组黏附细胞呈圆形或类圆形;黏附LST-R1细胞凋亡率分别为(3.46±0.89)%和(8.61±1.23)%。结论LST细胞黏附至细胞外基质后,膜表面galectin-1可促进黏附细胞凋亡增加,表明膜表面galectin-1可能参与了LST病变侧向扩散非侵袭性生长特性的形成。
Objective To observe the influence on adhered LST-R1 cell apoptosis by galectin-1 antibody inhibiting. Methods Laser cofocal scanning microscopy was employed to detect the expression of galectin-1 in LST-R1 cell. LST-R1 cells inhibited by galectin-1 antibody or not were seeded in the 48-well plates coated with collagen Ⅰ (20μg/well). The morphology of the adhesive LST-R1 cells was observed. Flow cytometer was employed to detect the adhered LST-R1 cell apoptosis. Results Laser cofocal scanning microscopy showed that galectin-1 expressed on the membrane of LST- R1 cells. The binging rate between galectin-1 and its antibody was (67.2 ± 6.73)%. In the galectin-1 antibody inhibiting group, more and more irregular angular adhesive cells appeared, but the adhesive cells in the control group were rounded or round-like. The apoptosis rats of adhesive LST-R1 cells in the antibody inhibiting group and the control group were (3.46±0.89)% and (8.61 ±1.23)% respectively(P 〈0.01). Conclusions Membrane galectin-1 may induce adherd LST-R1 cell apoptosis,which indicates membrane galectin-1 is associated with the noninvasive phenotype of laterally spreading tumor(LST) lesion possibly.
