摘要
目的:观察体外直流电场对人肺癌细胞系A549细胞的定向移行作用。方法:实验组将A549肺癌细胞暴露于强度分别为2V/cm、4V/cm、6V/cm及8V/cm的直流电场中,未暴露于电场的A549肺癌细胞作为对照。观察记录每5min该细胞移行的方向和变化,测量其移行的移行轨迹总长度、位移距离、速率及方向共3h。数据结果用SAS8.0及Image-ProPlus5.0软件处理。结果:未暴露于直流电场中的A549细胞在观察期间无明显移动,3h后细胞形态未见明显改变。而暴露于不同强度电场中的各组细胞则呈现有规律的两种变化:大部分细胞长轴趋向垂直于场线方向排列;细胞向阴极方向定向运动,且这种运动速度随电场增大而加快(P<0.05),细胞移行的方向系数(cosγ)随暴露时间的延长而增大(P<0.05)。结论:体外直流电场可诱导人肺癌细胞系A549细胞朝向阴极定向移行,此作用与电场强度有关,长时间暴露于电场中其移行速率与时间无关。
Objective: To study the effect of direct - current electric field on the directed migration of A549 cells. Methods: The A549 cells were cultured in DMEM + 10% FBS. Cells in experimental groups were exposed to the direct current electric field at 2V/cm, 4V/cm, 6V/cm and 8V/cm. Cells unexposed to the electric fields were used as control. Images were taken per 5 mins within 3 hrs and the correlated parameters were measured. Results: A549 cells unexposed to the electric field showed slightly random movements during 3 hrs. On the contrary, most of the cells in experimental groups aligned and elongated perpendicularly to the electric field lines and began to migrate to the cathode. This migration speed increased when voltage increased. The average directional cosine γ of cell migration was getting more with the time lapse, (P 〈 0.05 ). Conclusion : A549 cells have Galvanotaxis. Electric field is able to induce cathodal directed migration of the A549 cells. And this effect is correlated to the voltage other than the duration of the cells exposure in the field.
