活血清下中药对大鼠缺血-再灌注小肠凋亡蛋白酶活化因子-1表达的影响

Regulation of Traditional Chinese Medicine on Apoptotic Protease Activating Factor-1 in Rats with Intestinal Ischemia-reperfusion

摘要 目的:利用小肠缺血-再灌注动物模型,观察中药对小肠黏膜细胞凋亡的干预作用及对肠屏障的影响。方法:选用健康Wistar大鼠,随机分为正常对照组、模型组、承气合剂治疗组、复方丹参合剂治疗组、活血承气合剂(承气合剂+复方丹参合剂)治疗组。对动物模型再灌注6h、24h、72h时血浆内毒素、小肠黏膜上皮细胞凋亡情况、凋亡蛋白酶活化因子-1(Apaf-1)的表达分别进行检测和观察。结果:与模型组比较,各治疗组对于肠黏膜上皮细胞凋亡均有不同程度的抑制作用,其中活血承气治疗组各观察指标降低明显(P<0.01),效果最佳。结论:小肠缺血-再灌注早期肠黏膜上皮细胞凋亡是造成血中内毒素明显升高、肠屏障功能损伤的主要原因之一。活血清下法可以通过抑制Apaf-1表达来减弱肠黏膜上皮细胞的凋亡,从而达到降低内毒素水平,保护肠屏障功能的作用。 Objective To investigate the protective effect of traditional Chinese medicine on gut barrier injury induced by intestinal ischemia-reperfusion in rats. Methods The rats were divided into 5 groups including control group, model group, ChengQi(承气) prescription group, Fufang Danshen(复方丹参) prescription group and nuoXue- QingXia(活血清下) prescription group. After intestinal ischemia-reperfurion, serum endotoxin, apoptosis of enterocytes and the expression of apoptotic protease activating factor(Apaf- 1 ) were detected by microscopy at 6, 24 and 72 hours. Results The apoptosis of enterocytes and the complement of gut barrier were kept better in the group of HuoXueChengQi (活血承气) presciption than the others. Conclucion In the early stage of intestinal ischemia-reperfusion injury with the apoptosis of enterocytes intestinal mucosal barrier were subjected to severe damage. HuoXueQingXia prescription regulated the apoptotic protease activating factor-1 to reduce the level of serum endotoxin and protect the gut barrier.
