摘要 目的探讨皮肤鳞状细胞癌(SCC)中FHIT mRNA及其蛋白的表达和临床意义。方法采用多聚寡核苷酸原位杂交技术和免疫组织化学SP法,分别检测FHIT mRNA、FHIT蛋白在30例SCC组和21例正常皮肤组标本中的表达。结果在SCC组和正常皮肤组的标本中,FHIT mRNA、FHIT蛋白的阳性率分别为36.7%、43.3%和85.7%、95.2%。两组间比较差异均具有统计学意义(P<0.05);原位杂交与免疫组织化学两种实验结果具有一致性。结论FHIT在SCC组织中的低表达,可能对肿瘤的形成具有重要作用。也可能作为SCC早期诊断及治疗的理论依据。 Objective To study the expression of FHIT mRNA and its protein in dermal squamons cell carcinoma (SCC) and their clinical significance. Methods In situ hybridization and the SP immunohistochemical method were adopted to examine FHIT mRNA, and FHIT protein in 30 cases of squamons cell carcinomas of the skin and 21 normal skin cases. Results The positive rates of FHIT mRNA and FHIT protein were 36.7 %/85.7 % and 43.3 %/95.2 % (respectively) in SCC and normal skin cases. Comparing SCC and normal skin cases, the expressions of FHIT mRNA and FHIT protein showed significant differences ( P 〈 0.05 ). There was consistency of positive expression of FHIT mRNA and its protein. Conclusion Abnormal FHIT expression may play an important role in the genesis of SCC. Furthermore, examining the expression of FHIT may have potential benefit for early diagnosis and treatment of SCC.
