摘要
目的考察口服透明质酸(HA)的分布情况。方法1.同位素标记示踪法给药:给小鼠灌胃同位素标记的HA(125Ⅰ—HA),分别于药后不同时间点取样,研究HA在小鼠组织内的分布情况;2.连续口服给药:采用放射免疫法测定大鼠连续口服HA31d后皮肤中不同存在形式HA含量的变化情况。结果同位素标记示踪法给药表明,在小鼠13个受检组织中均有不同程度的放射性分布,1~4h脾脏的代谢率快于肝脏,睾丸与皮肤中代谢率明显慢于其它器官,药后12h内,38.7%的放射性从粪便排泄,18.6%从尿液排泄;连续口服给药表明,与对照组相比,大鼠31d后皮肤中HA总量有显著性升高,其中游离HA的含量有极显著性升高。结论口服HA被机体吸收后,能分布于机体的受检组织中,尤其可定位集中于皮肤组织。
Purpose To investigate the distribution of hyaluronic acid after oral administration. Methods 1. Isotope tracer labelling method: the samples of mice tissues were taken in different time after125 Ⅰ- HA was administered orally, then the radioactivity in sampling tissues was detected. 2. multiple-dose oral administration: the changes of contents of different types of HA in rats' skin were also determined by radioimmunoassay after taking orally HA for 31 days.Results The radioactivity was differently distributed in 13 sampling tissues. And there were many differences in the metabolic rate of each tissue in mice between 1 h and 4 h: the rate of spleen was faster than liver's, and the rate of testicle and skin was slower than the other tissues'. In 0-12 h, about 38.7 % of radioactivity was drained in feces, and 18.6% of radioactivity was drained in urine. In multiple-dose experiment, it was observed that there was a statistically significant increase of the content of total-HA in rat skin after taking orally HA for 31 days, especially there was a very significant increase of the content of free-HA in rat skin. Conclusion HA might be distributed in sampling tissues after oral administration and concentrated locationally in skin after being absorbed.
