局灶性脑缺血损伤后TNF-α、TGF-β_1表达的实验研究 被引量:2

摘要 目的:研究大鼠缺血性脑损伤后缺血区脑组织肿瘤坏死因α(TNF-α)和转化生长因子β1(TGF-β1)表达的变化,探讨葛根素对缺血性脑损伤的神经保护机制。方法:90只雄性SD大鼠,用线栓法建立大脑中动脉缺血再灌注模型,随机分为手术+葛根素组,手术组,假手术组(各30只)。每组又分五个不同时间点(6h,12h,24h,48h,72h),手术+葛根素组大鼠应用葛根素干预治疗,假手术组和手术组大鼠给予同等容量生理盐水对照。用SABA免疫组化染色观察各时间点缺血脑组织TNF-α、TGF-β1因子的表达。结果:与假手术组相比,手术组和手术+葛根素组脑缺血再灌注后TNF-α、TGF-β1蛋白的表达均显著增加(P<0.01)。手术+葛根素组脑缺血再灌注后不同时间点TNF-α的表达均较手术组显著下降(P<0.01,P<0.05);TGF-β1的表达均较手术组显著增加(P<0.01,P<0.05)。结论:葛根素可能通过抑制TNF-α因子的表达,诱导TGF-β1因子的表达,从而抑制过度的炎症反应来发挥神经保护作用。 Objective: To study the neuroprotective mechanism of puerarin on ischemic brain injured by observing the expression change of TNF-α,TGF-β1in ischemic brain area. Methods: 90 healthy male SD (Spraque-Dawiey) rats were built the animal model of brain medium-sized artery focal cerebral ischemia reperfusion with the improvement Zea-Longar patching methods and they were randomly divided into three groups: the sham operation group(n= 30); the operation group(n=30); the operation with puerarin interfered group (n= 30). There were five different observing time in every group(6h, 12h ,24h,48h, 72h). The operation with puerarin group rats were treated with puerarin solution, The sham operation and operation group rats were injected the same volume Sodium Chloride. To observe the expression of TNF-α,TGF-β1cytokines in different time with the method of SABA immunohisto chemical Staining in ischemic region. Results: Compared with the sham operation group, there were much more expression of the TNF-α,TGF-β1 in cerebral ischemic reperfusion brain tissue in the operation and the operation with puerarin interfered groups (P〈0. 01). Compared with the operation group , The expression of TNF-α was significantly decreased,while the expression of TGF-β1was improved rapidly in different time after cerebral ischemic-reperfusion in the operation with puerarin interfered group (P〈0.01or P〈0.05). Conclusion: The puerarin can restrain the expression of the TNF-α and induct the high level of the TGF-β1, thus the pueratin restrained the over Inflammatory reactiono to protect the brain tissue.
