高效液相色谱法测定血清中地西泮的浓度

Determination of diazepam sodium in serum by HPLC

目的建立血清中地西泮的高效液相色谱测定方法。方法以甲醇-水（70：30）为流动相；Kromasil C18（250mm×4．6mm5μm）柱为层析柱，以卡马西平为内标物，检测波长216nm。结果地西泮在0．1～20．0mg·L^-1浓度范围内具有良好的线性关系，平均回收率为97、67％，日内、日间RSD小于4．8％，最低检测浓度为20μg·L^-1。结论本方法简便可行，适合于地西泮的血药浓度测定。

Aim To eatablish an HPLC method for the determination of diazepam in serum. Methods Mobile phase consisted of metha- nol-water（70：30 ）. Kromasil C is （250 mm × 4.6 mm 5 μm） column was used,with carbamazepine as internal standard, and UV-detector wavelength was 216 nm. Results The linear range was between 0. 1 and 20.0 mg · L^-1. The average recovery was 97.67%, and RSD of within-day and between-day was less than 4.8%. Detection limits were above 20 ng · m L^-1. Conclusion This method is simple and feasible for the determination of diazepam in serum.
