摘要
通过对我国四大海域野生三疣梭子蟹(Portunus trituberculatus)9个群体线粒体DNA控制区基因片段进行扩增和测序,得到长度为530 bp的片段。分析结果表明:83条序列T、C、A、G4种核苷酸的平均含量分别为36.6%、16.1%、36.6%、10.7%。共检测到91个变异位点,其中缺失/插入位点2个,转换位点76个,颠换位点3个,转换、颠换同时存在位点10个。83个个体具有66种单倍型(hap lotype),单倍型比率为79.5%,对9个群体用MEGA3.1构建NJ分子树,聚类结果显示黄海、东海、渤海6个群体之间的相对遗传距离比较近,聚在了一起,南海3个群体相对遗传距离比较近,单独聚在一起。我国四大海域的野生三疣梭子蟹不同群体间有一定遗传分化。
Mitochondrial putative control region gene fiagments were amplified and sequenced from 9 wild stocks of Portunus trituberculatus fiom four seas of China. 530 bp long partial gene fiagments were obtained. The results showed that the A, T, G and C mean contents in those fiagments were 36.6% , 16.1% , 36.6% , 10.7%, respectively. There were 91 variation sites in those gene fiagments, which included 2 deletion/insert sites, 76 transition sites, 3 transvertion sites, 10 transition and transvertion sites. Comparisons of these 83 partial putative control region sequences revealed 66 mitochondrial haplotypes in P. trituberculatus, the haplotype rate was 79.5%. The molecular phylogenetic tree were constructed with NJ method using software MEGA 3.1. The genetic distance and cluster analysis showed that genetic distances were nearest among six stocks of Yellow Sea, East China Sea, Bohai Sea, while the genetic distances were nearest among three stocks of South China Sea. So these indicated that there were some genetic differentiation in different wild stocks of P. trituberculatus in China .
