摘要
目的探讨X射线照射后鼻咽癌细胞系HNE1产生多药耐药的机制。方法利用逆转录PCR检测照射前后HNE1细胞MDR1的基因表达变化,利用Western印迹检测照射前后MDR1基因编码蛋白P-gp的表达变化。结果鼻咽癌细胞系HNE1照射后MDR1基因表达增加,P-gp蛋白表达增加。结论X射线可以诱导鼻咽癌细胞系HNE1 MDR1基因的表达,这可能是照射后鼻咽癌细胞耐药的重要原因之一。
Objective To investigate the mechanism of muhidrug resistance gane in nasopharyngeal carcinoma cell(NPC) HNE1 after X-ray irradiation. Methods The expression of MDR1 gene and its protein P-gp were examined in HNE1 cell before and after X-ray irradiation by reverse transcripional polymerase chain reaction (RT-PCR), and western blotting, respectively. Result After X-ray irradiation, both the expression of MDR1 gene and its protein P-gp were increased. Conclusions X-ray irradiation may induce the expression of MDR1 gene in HNE1 cells, which may be play an important role in multidrug resistance after NPC irradiation.
