五味润僵汤治疗类风湿关节炎的实验研究

The Study on Therapy of WU-WEI-RUN-JIANG Decoction in Rheumatoid Arthritis

目的:研究五味润僵汤对类风湿性关节炎患者的疗效。方法:建立Ⅱ型胶原诱导关节炎大鼠模型。测量大鼠足肿胀度,计算肿胀率及关节炎指数。采用酶联免疫吸附测定法检测各组血清中的TNF-α和IL-1B表达的影响,并与模型组、正常组进行对比研究。结果:CIA大鼠有明显持续的局部关节肿胀的临床表现。给药后各组治疗均能减少大鼠足爪肿胀和关节指数,与模型组相比差异显著(P<0.01)。正常组的血清中TNF-α和IL-1B呈低表达,与模型组血清中的TNF-α和IL-1B的高表达有显著性的差异(P<0.01)。治疗组血清中TNF-α和IL-1B较接近于正常组的表达,与模型组差异有显著性(P<0.01)。结论:五味润僵汤治疗Ⅱ型胶原诱导性关节炎具有显著疗效,能够明显减轻大鼠的足爪肿胀,降低血清中TNF-α和IL-1B水平。

Objective： To study the curative effect of wu - wei- runjiang decoction in rheumatoid arthritis. Methods：The model of rats with rheumatoid arthritis induced by type Ⅱ collagen was established. The feet swelling degrees of rats were measured, and then calculated the swelling rate. Enzyme - linked immunosorbent assay technique has been used to detect cytokines TNF -α and IL - 1B in blood serum of CIA rats. The above results were compared with those of model group and normal group. Result：The CIA rats had obvious and persistent arthrocele clinical manifestation. The treating groups had more obvious effects than model group on reducing foot swelling and arthritis index（P 〈 0.01）. The expression of TNF-α and IL- 1B in the blood serum were lower in normal group than those in model group（P〈0.01）. The expressions of TNF-α and IL- 1B in treating groups were similar to those in normal group and different from those in model group（P 〈 0.01）. Conclusion： Wu- wei - run-jiang decoction has significant effect in rheumatoid arthritis induced by type IT collagen . It can reduce feet swelling of CIA rats , decrease TNF -α and IL - 1B in blood serum.
